Abstract

This study aims to determine the biometric and morphometric differences of Kacang, Ettawa (PE) and Boer buck spermatozoa. Semen collected by an artificial vagina. One part of fresh semen is diluted with ten parts of physiological (v/v) NaCl. Fixation of slides was performed by mixing one drop of dilute semen with one drop of Eosin-nigrosin solution, homogenized and dried on a hot plate at 40°C for several minutes. Biometry and morphometry of spermatozoa were measured using a 400x magnification phase contrast microscope (CarlZeis, Germany). One slide for each buck semen samples was made and 10 samples of live spermatozoa were taken from each slide, so that the spermatozoa samples for each type of buck was 50 spermatozoa. The significance between variables was tested using the T-test with independent samples test with a significance level of 0.05. The results showed that there was no significant difference in the biometric variables of the three types of bucks, except that the concentration, mass motility and individual motility of PE buck spermatozoa were significantly lower than Boer and Kacang bucks. The head length of the spermatozoa of Kacang, Etawah and Boer was not significantly different. In contrast, the head width of the Boer buck spermatozoa tended to be wider but not significantly different than the Kacang and PE bucks. Meanwhile, the spermatozoa tail length of Boer buck was significantly shorter than the Kacang and PE buck. In conclusion, the spermatozoa of PE and Boer buck sperm were not significantly different. It is recommended that the results be more accurate, measurement of spermatozoa morphometry should be carried out on spermatozoa with the same chromosome, namely X or Y only.berbeda nyata dibanding kambing Kacang dan kambing PE. Sedangkan panjang ekor spermatozoa kambing Boer secara signifikan lebih pendek dibanding kambing Kacang dan kambing PE. Disimpulkan, Morfomerti spermatozoa kambing Kacang PE dan Boer tidak berbeda nyata. Disarankan supaya hasilnya lebih akurat, pengukuran morfomerti spermatozoa hendaknya dilakukan pada spermatozoa dengan kromosom yang sama yaitu X atau Y saja
