Abstract

The aim of this study was to compare yield of bovine embryos obtained from several number of culture conditions and culture systems. Ovaries were collected from a local slaughterhouse and transported to the laboratory in PBS. Ovarian follicles (2-6 mm) were aspirated within 3 h of slaughter, using TCM-199 supplemented with 10% fetal calf serum. The oocytes were matured in TCM 199 under paraffin oil and incubated at 38ºC in a humidified atmosphere in air for 24 to 26 hours. Folicullar bovine cumulus oocyte complexes were matured in vitro and in vitro fertilization was carried out with the frozen semen of Bali cattle bull. Forty eight hours after in vitro fertilization. Under cell-free culture condition (modified Synthetic Oviduct Fluid/mSOF supplemented with 10% FCS) 48 h post fertilization, rate of development day 10 blastocysts of embryos cultured in small groups (1-5 per drop) was lower than that in large group (8 versus 25%; P<0.01). There was no group effect when culture system allowing the progressive development of cumulus cells. This shows that somatic cells improve the development of single cultured bovine embryos up to the blastocyst stage; and cooperation between embryos could replace the effect of co-culture either on yield of blastocysts. Blastocysts appeared significantly earlier in mSOF than in co-culture with cumulus cell in TCM 199 and bovine oviduct epithelial cell (BOEC) in TCM 199 (detection of the blastocysts: 6.7±0.2 d post insemination with mSOF versus 7.2±0.2 d post insemination with cumulus cell and 7.5±0.2 d post insemination in BOEC P<0.01). Results show that there was improved cumulus cell development by incubating single oocyte than that of 5 or 10. Co culture media: SOF, cumulus cells and Bovine Oviduct Epithelial Cell (BOEC) in TCM 199 produced blastocyst by incubating oocyte for 6.7±0.2 days, 7.2±0.2 days and 7.5±0.1 days following sperm insemination. It is concluded that development of cumulus cells improve by incubating less oocyte and co culture media SOF produce blastocyst earlier than that of cumulus cell and oviduct cells co culture
