Abstract

Diabetes mellitus is a disease with high prevalence in Indonesia. Due to the side effects and expensive prices of consuming synthetic antidiabetic drugs are increased many researches on exploring of natural substances as antidiabetic, one of them is Brucea javanica (L) Merr. Brucea javanica (L) Merr can reduce blood glucose level. The mechanism of action of Brucea javanica (L) Merr in reducing blood glucose level can be determined by measuring glycogen levels in the liver and muscles. The aim of this study was to determine the effect of Brucea javanica (L) Merr seeds suspensed on glycogen levels in the liver and muscles of diabetic rats. This study used five groups; positive control (glibenclamide 1.35 mg/kgbw), normal control, negative control (CMC-Na 0.5%), and treatment with dose of 25 mg/kgbw and 50 mg/kgbw. All groups (except normal control) were induced with alloxan 125 mg/kgbw intraperitonially. Glycogen levels of liver and muscles in all treatments were measured following the anthrone method by using UV-Vis spectrophotometric instrument. The glycogen level of each treatment was analyzed statistically by ANOVA using SPSS 16. The results of this study were suspensed doses of 25 mg/kgbw and 50 mg/kgbw were able to increase glycogen levels in the liver and muscles of diabetic rats. The increase in the dose of 25 mg/kgbw was higher than the dose of 50 mg/kgbw, with an increased percentage of 131.58% in active muscle, 258.6% in passive muscle, and 217.18% in the liver. Statistical analysis showed that glycogen levels in the suspensed group significantly different (p<0.05) compared to the normal, positive, and negative controls.
