Abstract

The purpose of this study was to analyze the profile of Bali cattle (Bos sondaicus) salivary proteins extracted with or without Polyethylene Glycol (PEG). The study was conducted at the Immunobiology Laboratory of the Faculty of Mathematics and Sciences, University of Mataram. The materials used were saliva collected from 3 Bali cattle. Saliva collected was extracted using PEG 6000 at a concentration of 10 %, 20% and 30% followed by the collection of supernatant and precipitate. The precipitate was solubilized with 250 µL of PBS, and the supernatant was collected in micro tubes and were dialyzed using physiological saline. Salivary protein concentration was measured using Bradford reagent. Salivary protein profiles were analyzed using 1D SDS-PAGE method. The results obtained demonstrate that the salivary protein bands of untreated samples are around 5-6 bands with a molecular weight range of 14-240 kDa. Whilst those extracted using 10%, 20% and 30% PEG showed around 4-6 visible bands with a molecular weight (MW) range of 14-240 kDa, 4-10 bands with a MW range of 7-240 kDa and 4-5 bands a MW range of 14-240 kDa, respectively.
