Abstract

The study entitled "Profile of Saliva Proteins of Bali Cattle (Bossondaicus) Extracted Using Cuprum Sulfate on 1-D SDS-PAGE" has been carried out in the Immunobiology Laboratory of the Faculty of Mathematics, University of Mataram (Unram) from October 2013 to January 2014 in order to determine the profile of Bali cattle’s saliva proteins extracted using cuprum sulfate (CuSO4). The materials used in this study were the saliva of Bali cattle collected from three Bali cows of 2.5 years old (coded A, B and C), with a healthy body condition and were not being pregnant. The cows were kept in Teaching Farm of Faculty of Animal Science, Unram, fed with King Grassas needed, and water was given ad libitum. Cuprum sulfate (CuSO4) used for the extraction of salivary proteins consisted of three final concentrations (v/v) namely 2.5, 5 and 10 mΜ. The results of the extractions consisted of the precipitate and the supernatant from each treatment were dialyzed and analyzed using a single dimension (1-D) SDS-PAGE. The qualitative analysis showed that there were variations in the patterns of salivary protein band in both treatments and controls. The number of protein bands in the control of Bali cattle A, B, and C were 10, 8 and 10 respectively, with molecular weight range of 14-240 kDa. Extraction using 2.5 mΜ CuSO4 resulted in similar results in all samples. There were9 protein bands appeared on the precipitate and the supernatant with a molecular weight range of 14-240 kDa. The 5 mΜCuSO4 gave a variation protein bands ranged from 2-10 protein bands in the precipitate and 9-10 protein bands in the supernatant with molecular weight range of 14-240 kDa, whereas the 10 mΜ treatment gave a protein patterns with the ranged of 9-10 protein bands in the precipitate and 2 -9 protein bands in the supernatants, with molecular weight of 14-240 kDa. Furthermore, the results of the extraction of the three Bali cattle saliva proteins using CuSO4showed the present of a predominant protein band with a molecular weight of 43.3 kDa. From this study it can be concluded that CuSO4 can be used to isolate salivary protein of Bali cattle with a molecular weight of 43.3 kDa.
