Abstract

Polymerase Chain Reaction (PCR) is a well-known molecular technique for DNA amplification and is widely implemented in diagnose of genetic disorders, identification of pathogen, DNA sequencing, forensic analysis and in the field of agriculture. The basic principle of PCR is integrated in the third year of High School students. Nevertheless, few high school teachers have been trained regarding this technique. Therefore, we feel the need to train high school teachers with the sufficient knowledge and skills regarding PCR hence would equip them with the necessary understanding of PCR to be implemented in the classroom level. As many as 12 teachers representing 12 High Schools around Mataram took part in this training The training comprises of two main parts, the first session provides teachers with basic understanding of genetics and PCR through means of classical presentation and emphasised with video illustration. The second session is a mix of hands on practice and demonstration of PCR technique in the laboratory. In this session, teachers have the opportunity to familiarize themselves with each steps of PCR from preparing the PCR mix to identification of the PCR results through gel electrophoresis. We observed the knowledge of teachers before and after the training. Average pre-test results were 44,4% (ranging from 10% to 70%) and the average post test result was 83.33% (ranging from 70% to 100%). Reflecting on the positive feedbacks from teachers it is necessary to conduct similar training for other high school teachers that have not yet taken part in this training.
